The Gunners, who have won the trophy twice, have home advantage in the clash, which will take place in the week beginning November 4th.
Holders Blackburn begin their defence with a home tie against First Division Walsall.
Last season's beaten finalists Tottenham travel to Burnley, while six-times winners Liverpool face Southampton.
There is only one other all Premiership contest, with Newcastle meeting Everton at St James' Park.
Manchester United are at home to Leicester, while Oxford United were rewarded for their midweek victory at Charlton with a home clash against five-times winners Aston Villa.
The other Third Division survivors, Bury, will play Fulham in the third round, while Second Division Wigan, conquerors of West Brom, have home advantage in their clash with Manchester City.
Sheffield United will meet Leeds in a Yorkshire derby at Bramall Lane, while 1998 winners Chelsea will play Gillingham at home.
