And ACT Brumbies flanker Owen Finegan has been recalled after missing Australia's victory over New Zealand Maori earlier this month through suspension.
Back-row man Matt Cockbain (skin infection) and wing Joe Roff (ankle injury) also sat out that game, but have now regained full fitness and return to the fray.
Both Queensland loosehead Stiles and ACT tighthead Darwin are poised to make their Test debuts in Brisbane, although Darwin could start as a substitute behind Glenn Panoho.
Unlike Lions coach Graham Henry, Macqueen has experienced no major injury setbacks during the build-up to Saturday's clash.
